Peter Claver Cullen was born in Montreal on July 28, 1941. As a kid, he spent time on his relatives' farm. He completed his education at the National Theatre School of Canada in 1963. Cullen worked as an announcer at CKGM (a Montreal radio station) and played an astronaut in a CFCS children's series, The Buddies. He then moved to Hollywood and joined The Smothers Brothers Comedy Hour, Sonny & Cher, and The Hudson Brothers Razzle Dazzle Comedy Show.

In his six-decade-long career, Cullen has voiced several characters, including Eeyore in Winnie the Pooh. However, his most popular role would be Optimus Prime.
